=== Story === For the first year of development, the team focused mostly on the gameplay without narrative structure. Playtesters found the game fun but asked about what these test chambers were leading towards. This prompted the team to come up with a narrative for Portal. The team worked with Marc Laidlaw, the writer of the Valve's Half-Life series, to fit Portal into the Half-Life universe. This was done in part because of the project's limited art resources; instead of creating art assets for Portal, the team reused the Half-Life 2 assets. Laidlaw opposed the crossover, feeling it "made both universes smaller", and said later: "I just had to react as gracefully as I could to the fact that it was going there without me. It didn't make any sense except from a resource-restricted point of view." Valve hired Erik Wolpaw and Chet Faliszek to write Portal. Wolpaw felt that the constraints improved the game. The concept of a computer AI guiding the player through experimental facilities to test the portal gun was arrived at early in the writing process. === Effects on humans and animals === Ethylene oxide is an alkylating agent; it has irritating, sensitizing, and narcotic effects. Chronic exposure to ethylene oxide is also mutagenic. The International Agency for Research on Cancer classifies ethylene oxide into group 1, meaning it is a proven carcinogen. Ethylene oxide is classified as a class 2 carcinogen by the German MAK commission and as a class A2 carcinogen by the ACGIH. A 2003 study of 7,576 women exposed while at work in commercial sterilization facilities in the US suggests ethylene oxide is associated with breast cancer incidence. A 2004 follow up study analyzing 18,235 men and women workers exposed to ethylene oxide from 1987 to 1998 concluded "There was little evidence of any excess cancer mortality for the cohort as a whole, with the exception of bone cancer based on small numbers. Positive exposure-response trends for lymphoid tumors were found for males only. Reasons for the sex specificity of this effect are not known. There was also some evidence of a positive exposure-response for breast cancer mortality." An increased incidence of brain tumors and mononuclear cell leukemia was found in rats that had inhaled ethylene oxide at concentrations of 10, 33 or 100 mL/m3 (0.0100, 0.0329 or 0.0997 imp fl oz/cu ft) over a period of two years. == Disorder and disease == Intrinsically unstructured proteins have been implicated in a number of diseases. Aggregation of misfolded proteins is the cause of many synucleinopathies and toxicity as those proteins start binding to each other randomly and can lead to cancer or cardiovascular diseases. Thereby, misfolding can happen spontaneously because millions of copies of proteins are made during the lifetime of an organism. The aggregation of the intrinsically unstructured protein α-synuclein is thought to be responsible. The structural flexibility of this protein together with its susceptibility to modification in the cell leads to misfolding and aggregation. Genetics, oxidative and nitrative stress as well as mitochondrial impairment impact the structural flexibility of the unstructured α-synuclein protein and associated disease mechanisms. Many key tumour suppressors have large intrinsically unstructured regions, for example p53 and BRCA1. These regions of the proteins are responsible for mediating many of their interactions. Taking the cell's native defense mechanisms as a model drugs can be developed, trying to block the place of noxious substrates and inhibiting them, and thus counteracting the disease.
=== Dentistry === As of 2019, the use of hydroxyapatite, or its synthetically manufactured form, nano-hydroxyapatite, is not yet common practice. == Release and development == The first season of the series was greenlit by Imagicomm Entertainment in March 2023, and was filmed in Charlotte, North Carolina. The official trailer for the series was released on YouTube in February 2024. The first episode aired on April 7, 2024. The second season completed filming in 2025, with both INSP and Universal TV announcing they would air the second season in 2026. In May 2026, the return of Blue Ridge to INSP was confirmed with the first episode of the second season airing July 19, 2026. Pharmacometabolomics, also known as pharmacometabonomics, is a field which stems from metabolomics, the quantification and analysis of metabolites produced by the body. It refers to the direct measurement of metabolites in an individual's bodily fluids, in order to predict or evaluate the metabolism of pharmaceutical compounds, and to better understand the pharmacokinetic profile of a drug. Alternatively, pharmacometabolomics can be applied to measure metabolite levels following the administration of a pharmaceutical compound, in order to monitor the effects of the compound on certain metabolic pathways(pharmacodynamics). This provides detailed mapping of drug effects on metabolism and the pathways that are implicated in mechanism of variation of response to treatment. In addition, the metabolic profile of an individual at baseline (metabotype) provides information about how individuals respond to treatment and highlights heterogeneity within a disease state. All three approaches require the quantification of metabolites found in bodily fluids and tissue, such as blood or urine, and can be used in the assessment of pharmaceutical treatment options for numerous disease states.
=== Non-medical use === Combining a muscle relaxant like carisoprodol with opioids and benzodiazepines is known as "The Holy Trinity" as it has been reported to increase the power of the "high". Recreational users of carisoprodol usually seek its potentially heavy sedating, relaxant, and anxiolytic effects. Because of its potentiating effects on narcotics, it is often used in conjunction with opioids. Carisoprodol is not tested for on standard drug testing screens, although tests for it do exist. On 26 March 2010 the DEA issued a Notice of Hearing on a proposal for placing carisoprodol in schedule IV of the Controlled Substances Act, later confirming its classification under schedule IV. Carisoprodol is sometimes mixed with date rape drugs.

== Medical uses == Setmelanotide is indicated for chronic weight management (weight loss and weight maintenance for at least one year) in people six years and older with obesity due to three rare genetic conditions: pro-opiomelanocortin (POMC) deficiency, proprotein subtilisin/kexin type 1 (PCSK1) deficiency, and leptin receptor (LEPR) deficiency confirmed by genetic testing demonstrating variants in POMC, PCSK1, or LEPR genes considered pathogenic (causing disease), likely pathogenic, or of uncertain significance. Setmelanotide is the first FDA-approved treatment for these genetic conditions. Setmelanotide is not approved for obesity due to suspected POMC, PCSK1, or LEPR deficiency with variants classified as benign (not causing disease) or likely benign or other types of obesity, including obesity associated with other genetic syndromes and general (polygenic) obesity.
== Career == After his PhD, Wüthrich continued postdoctoral research with Fallab for a short time before leaving to work at the University of California, Berkeley for two years from 1965 with Robert E. Connick. That was followed by a stint working with Robert G. Shulman at the Bell Telephone Laboratories in Murray Hill, New Jersey from 1967 to 1969. Wüthrich returned to Switzerland, to Zürich, in 1969, where he began his career there at the ETH Zürich, rising to Professor of Biophysics by 1980. He currently maintains a laboratory at the ETH Zürich, at The Scripps Research Institute, in La Jolla, California and at the iHuman Institute of ShanghaiTech University. He has also been a visiting professor at the University of Edinburgh (1997–2000), the Chinese University of Hong Kong (where he was an Honorary Professor) and Yonsei University. During his graduate studies Wüthrich started out working with electron paramagnetic resonance spectroscopy, and the subject of his PhD thesis was "the catalytic activity of copper compounds in autoxidation reactions". During his time as a postdoc in Berkeley he began working with the newly developed and related technique of nuclear magnetic resonance spectroscopy to study the hydration of metal complexes. === Reunited Germany, 1990–present === East Germany had originally consisted of five states (i.e., Brandenburg, Mecklenburg-Vorpommern, Saxony, Saxony-Anhalt, and Thuringia). In 1952, these states were abolished and the East was divided into 14 administrative districts called Bezirke. Soviet-controlled East Berlin – despite officially having the same status as West Berlin – was declared East Germany's capital and its 15th district. The debate on territorial revision restarted shortly before German reunification. While academics (Rutz and others) and politicians (Gobrecht) suggested introducing only two, three, or four states in East Germany, legislation reconstituted the East German states in an arrangement similar to that which they had had before 1952, as the five "New States" on 3 October 1990. The former district of East Berlin joined West Berlin to form the new state of Berlin. Henceforth, the 10 "old states", plus 5 "new states", plus the new state of Berlin, add up to the current 16 states of Germany. After reunification, the constitution was amended to state that the citizens of the 16 states had successfully achieved the unity of Germany in free self-determination and that the West German constitution thus applied to the entire German people. Article 23, which had allowed "any other parts of Germany" to join, was rephrased. It had been used in 1957 to reintegrate the Saar Protectorate as the Saarland into the Federal Republic, and this was used as a model for German reunification in 1990.
== Effects on pharmacokinetics == Those experiencing poverty-related infectious diseases (PRDs) such as malaria and tuberculosis are also likely to be malnourished. Malnutrition can affect the pharmacokinetics of various drugs used to treat PRDs by changing a drug's bioavailability, distribution, and elimination. To optimize the treatment of those diseases, there needs to be more research into how severe malnutrition, specifically kwashiorkor, can affect treatment response.
=== α2-Adrenergic receptor agonists === α2-Adrenergic receptor agonists like clonidine can improve sleep and may be useful in the treatment of insomnia. An example of this is in the treatment of insomnia in children and adolescents with attention deficit hyperactivity disorder (ADHD), for instance due stimulant therapy. Similarly to clonidine, the α2-adrenergic receptor agonist dexmedetomidine has sedative and hypnotic effects and is used to produce sedation in hospital settings. The sleep induced by dexmedetomidine is said to closely resemble natural sleep. The selective α2A-adrenergic receptor agonist tasipimidine (ODM-105) is under development for the treatment of insomnia and is in phase 2 clinical trials for this indication as of October 2024. α2-Adrenergic receptor agonists can produce hypotension and bradycardia as side effects, which has limited their use. Activation of the α2A-adrenergic receptor is thought to be responsible for most of the physiological effects of the α2-adrenergic receptors, including hypotension. On the other hand, the preferential α2A-adrenergic receptor agonist guanfacine appears to show less sedation and hypotension than clonidine. Tizanidine has been used off-label for sleep similarly to clonidine, but likewise shows less hypotensive potency in comparison.
