Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
81. ArXiv [Preprint]. 2026 Jul 29:arXiv:2605.17186v2. Operator splitting for exploiting linear-rate closure in solving infinite ODE hierarchies. Chang JC. We introduce an operator-splitting method for infinite hierarchies of linear ordinary differential equations (ODEs) indexed by nonnegative integers. When the coupling coefficients depend linearly on the count index, an exact transformation closes the equations on finite count-index windows without an upper-boundary value. For more general hierarchies, Strang splitting applies the linear-rate closure during the linear-rate substeps and a conventional capped solver to the remainder. We derive the closure from generating functions and the method of characteristics and extend it to multi-indexed systems. The derivation requires neither positivity nor mass conservation, so it applies to a wider class of systems than the examplar stochastic models presented here. We discuss branching processes, stochastic predator-prey dynamics, the Schlögl chemical kinetics model, and a telegraph model for gene expression. Through numerical experiments and computational cost analyses we demonstrate that our operator splitting method is typically advantageous for solving large scale systems in terms of memory usage and computational time, while retaining accuracy competitive with finite state projection (FSP) methods. According to the United States Geological Survey (USGS), 10,200 tonnes of bismuth were produced worldwide by mining and 17,100 tonnes by refining in 2016. Since then, USGS does not provide mining data for bismuth, considering them unreliable. Globally, bismuth is mostly produced by refining, as a byproduct of extraction of other metals such as lead, copper, tin, molybdenum, and tungsten, though the refining-to-mining ratio depends on the country. Bismuth travels in crude lead bullion (which can contain up to 10% bismuth) through several stages of refining, until it is removed by the Kroll-Betterton process, which separates the impurities as slag, or the electrolytic Betts process. Bismuth behaves similarly with another of its major metals, copper. The raw bismuth metal from both processes contains still considerable amounts of other metals, foremost lead. By reacting the molten mixture with chlorine gas, the metals are converted to their chlorides, while bismuth remains unchanged. Impurities can also be removed by various other methods, for example, with fluxes and treatments yielding high-purity bismuth metal (over 99% Bi).

Amino acids are a key nutrient in ecosystems. Some are essential to animals, meaning that these organisms cannot synthesize them de novo. Instead, animals rely on their diet to acquire these molecules, creating strong interdependencies between animals and organisms with complete amino acid synthesis capabilities. In a study of bacteria and archaea at Antarctica's McMurdo Dry Valleys, the distribution of 13C between their amino acids reflected the biosynthetic pathways employed by these organisms. Autotrophs and heterotrophs had distinct isotopic fingerprints, as did organisms that employed alternatives to the citric acid cycle to ferment or produce acetate. Plants, fungi, and bacteria are also distinguishable by their amino acid carbon isotopes. The compositions of the essential amino acids, which have more complex biosynthetic pathways, are particularly informative. Lysine, isoleucine, leucine, threonine, and valine all had significantly different δ13C values between at least two of these groups. The fungi and bacteria in this study were grown on amino acid-free media to ensure that all the amino acids were synthesized by the organisms of interest. Bacteria and fungi can also scavenge amino acids from the environment, complicating the interpretation of data from field samples. Nevertheless, researchers have successfully used these differences to identify the sources of amino acids in food webs. Terrestrial and marine producers in a mangrove forest had different patterns of 13C enrichment in their amino acids.

A 2006 United States Department of Agriculture report found the adoption of genetically engineered (GE) soy, corn and cotton reduced the amount of pesticides used overall, but did result in a slightly greater amount of herbicides used for soy specifically. The use of GE soy was also associated with greater conservation tillage, indirectly leading to better soil conservation, as well as increased income from off-farming sources due to the greater ease with which the crops can be managed. Though the overall estimated benefits of the adoption of GE soybeans in the United States was $310 million, the majority of this benefit was experienced by the companies selling the seeds (40%), followed by biotechnology firms (28%) and farmers (20%). The patent on glyphosate-tolerant soybeans expired in 2014, so benefits can be expected to shift.
"There's no way Rush will ever exist again because Neil's not here to be a part of it... that's not to say that we can't do other things and we can't do things that benefit our communities and all of that. I have lots of plans for that sort of thing that don't necessarily include Geddy. I get asked this all the time—are we gonna do this, or are we gonna do that? Who knows? All I know is we still love each other and we're still very, very good friends, and we always will be."
== History == Elected county councils were established in 1889 under the Local Government Act 1888, taking over administrative functions previously carried out by unelected magistrates at the quarter sessions. The boroughs of Gateshead, South Shields and Sunderland were considered large enough to provide their own county-level services and so they were made county boroughs, independent from Durham County Council. The county council was elected by and provided services to the rest of the county, which area was termed the administrative county. Additional county boroughs were later created at West Hartlepool in 1902 and Darlington in 1915. In 1967 West Hartlepool merged with the neighbouring borough of Hartlepool (which had just covered the old town), with the enlarged county borough thereafter being called Hartlepool. Stockton-on-Tees, Billingham and surrounding areas were removed from the administrative county in 1968 to become part of the County Borough of Teesside.

=== Blood and lymphatic vessels === The prostate receives blood through the inferior vesical artery, internal pudendal artery, and middle rectal arteries. These vessels enter the prostate on its outer posterior surface where it meets the bladder, and travel forward to the apex of the prostate. Both the inferior vesical and the middle rectal arteries often arise together directly from the internal iliac arteries. On entering the bladder, the inferior vesical artery splits into a urethral branch, supplying the urethral prostate; and a capsular branch, which travels around the capsule and has smaller branches, which perforate into the prostate. The veins of the prostate form a network – the prostatic venous plexus, primarily around its front and outer surface. This network also receives blood from the deep dorsal vein of the penis, and is connected via branches to the vesical plexus and internal pudendal veins. Veins drain into the vesical and then internal iliac veins. The lymphatic drainage of the prostate depends on the positioning of the area. Vessels surrounding the vas deferens, some of the vessels in the seminal vesicle, and a vessel from the posterior surface of the prostate drain into the external iliac lymph nodes. Some of the seminal vesicle vessels, prostatic vessels, and vessels from the anterior prostate drain into internal iliac lymph nodes. Vessels of the prostate itself also drain into the obturator and sacral lymph nodes.
==== Seeds ==== Lotus seeds are mostly oval or spherical, with sizes varying according to varieties. They are generally 1.2–1.8 cm long, with diameters ranging from 0.8 to 1.4 cm and a weight of 1.1–1.4 g. After lotus seeds have been decorticated and peeled, they are edible and rich in nutrients and can be dried for storage. Their nutritional values can differ due to culture environments and varieties. Not only do these seeds contain proteins of high quality and are rich in a variety of essential amino acids including high contents of albumin (42%) and globulin (27%), they also contain unsaturated fatty acids, carbohydrates, vitamins, calcium, iron, zinc, phosphorus and other trace elements. They also provide water-soluble polysaccharides, alkaloids, flavonoids, superoxide dismutase, and other bioactive components. Lotus seeds also contain particularly large amounts of vitamins, including VB1, VB2, VB6 and Vitamin E. The functional components (polyphenols, protein, polysaccharides) in N. nucifera seeds can help combat high blood pressure, diabetes, and gallstones. After lotus seed germination, crude protein and fat levels in the endosperm significantly increase. It is therefore an important method to enhance its nutritional quality.
